Research Interests
I am interested in the work for resistant resource screening and the mechanism between pathogen and host by scanning electron microscope and some molecular techniques such as enzyme assay, determination of biocontrol compound, Real-time PCR, cDNA-AFLP and RNAseq for transcriptional analysis, and 2D gels for proteomics analysis.
Major Projects
For master degree
- Isolated a new protease inhibitor gene from the nematodes symbiotic bacteria Xenorhabdus bovienii and revealed the insecticidal bioactivity and mechanism against agricultural insect pests (Zeng F., Xue R. , et al., 2012, Pest Management Science; Xue R. F. et al., 2009, Biotechnology Bulletin (in Chinese); Jin D.J., Xue R. F. , et al., 2012, Chinese Journal of Biological Control (in Chinese) and 1 manuscript in preparation).
- A new protease inhibitor gene designed as PIN1 was cloned from Xenorhabdus bovienii ;
- The recombinant protein expressed in E. coli was purificated by affinity chromatography;
- The insecticidal bioactivity and mechanism were determined against several important agricultural insect pests (aphids, green leaf bugs and whitefly) using the insect bioassay, measurement of enzyme activity and establishment of transgenic tobacoo.
- Isolated a new toxin protein gene from Xenorhabdus bovienii and reveal the insecticidal bioactivity and mechanism (1 manuscript in preparation).
- A new toxin protein gene designed as Xbtp-1 was cloned from Xenorhabdus bovienii ;
- The recombinant protein expressed in E. coli was purificated by affinity chromatography;
- The insecticidal bioactivity was determined against Helicoverpa armigera using the insect bioassay and measurement of digestive enzyme activity in midgut of insects;
- Illumination of the insecticidal mechanism of Xbtp-1 showed that the Xbtp-1 recombinant protein enhanced the tissue degradation of the insect midgut by scanning electron microscopy.
For Ph.D. degree
- Isolated and identified Fusarium wilt pathogens and screened the resistant cultivar of common bean for Fusarium oxysporum f. sp. phaseoli (Fop).
- 70 isolates of F. oxysporum f. sp. phaseoli were isolated and identified by phenotype, genotype (self-designed primer) and pathogenicity test, and an isolate of Fusarium oxysporum f. sp. phaseoli — Fop-DM01 exhibited strong virulence were used for screening of the resistant cultivars;
- 362 cultivars of common bean were screened, and one cultivar (260205) showed stable resistance to Fop-DM01 isolate.
- Designed a specific primer of F. oxysporum f. sp. phaseoli and distinguished the resistance levels of different bean cultivars by Real-time PCR. ( Xue R. F. , et al., 2012, Acta Agronomica Sinica)
- A pair of specific primers was designed depended on a new transcriptional factor gene ( ftf1 genes) from Fop;
- The capacity of pathogen colonization on different cultivars was distinguished by R eal time-PCR and verified by greenhouse experiment.
- The study of the interaction between common bean and the pathogen Fop-DM01 isolate . (1 manuscript in preparation)
- The organizational and celluar structure of infected roots and stems in susceptible cultivar BRB-130 which displayed by scanning electron micrographs and transmission electron micrographs degraded more seriously than that in resistant cultivar 260205.;
- The activity of the plant defence-related emzymes, phenylalanine ammonia lyase (PAL) and peroxidase (POX), and the content of H 2 O 2 and O 2- as the signalling molecule of plant defence pathway in roots and stems of 260205 was higher than BRB-130;
- Differentially expressed defence-related genes between the infected resistant 260205 and infected susceptibe BRB-130 were revealed by RNAseq and cDNA-AFLP analysis;
- Proteomics analysis using 2D gels to discover the key proteins in the plant defence to the pathogen.
- Cloning of two defence-responsed genes from the resistant cultivar 260205 named PvCaM1 encoding calmodulin and PvPOX1 encoding secretory peroxidase, separately. ( Xue R. F. , et al., 2012, Acta Agronomica Sinica (in Chinese) and 1 manuscript in preparation)
- Cloning and analysis of the full-length cDNA and DNA of the two genes by Race and RT-PCR;
- The expression level of the two genes in different organs and in response to biotic and abiotic stress by real time-PCR analysis (e.g. pathogen attack, wounding, cold, salt, drought and plant hormone);
- Subcellular localization using GFP as a marker showed PvCaM1 was located in cytoplasm and PvPOX1 was secreted to extracellular matrix .
- Resistance response to Fusarium oxysporum f. sp. phaseoli in common bean ( Phaseolus vulgaris L.) induced by salicylic acid (SA). ( Xue R. F. , et al., 2013, Journal of Plant Growth Regulation)
- The resistance of susceptible cultivar BRB-130 was induced by SA treatment;
- HPLC analysis indicated the content of free and conjugated SA in roots increased after SA treatment ;
- PAL and POX activities in roots were both enhanced by SA treatment;
- H 2 O 2 and O 2- content in roots increased after SA treatment;
- The capacity of pathogen colonization in roots decreased after SA treatments detected by real time-PCR.
